EFFECT OF LOW ENDOGENOUS BRAIN-DERIVED NEUROTROPHIC FACTORLEVELS ON STRIATAL DOPAMINE DYNAMICSbyFRANCIS KABUI MAINADecember 2011Advisor: Dr. Tiffany A. MathewsMajor: Chemistry (Analytical)Degree: Doctor of PhilosophyMany neurological diseases and disorders are a result of alterations withneurotransmitters, neuromodulators, and/or proteins. Specifically, one protein that hasbeen linked to numerous neurological diseases and disorders such as Parkinson’sdisease, Huntington’s disease, addiction, attention deficit hyperactivity disorder (ADHD),and depression is brain-derived neurotrophic factor (BDNF). As a trophic factor, BDNFrole is to assist in the growth, survival, and differentiation of neurons. However, there isincreasing evidence that BDNF may mediate neurotransmitter dynamics. Our goalswere to understand how endogenous BDNF levels and aging modulate the dopamine(DA) dynamics in the mouse striatum. Two complementary neurochemical techniques, slice (in vitro) fast scan cyclic voltammetry (FSCV) and in vivo microdialysis were used to characterize striatal DA dynamics in wildtype and BDNF heterozygous mutant(BDNF) mice.Overall, our findings highlight that (1) we were able to develop a reliable FSCVassay to characterize DA release-regulating D2 and D3 autoreceptors functionality inthe striatum of wildtype (C57BL/6) mice, (2) BDNF mice appear to be

Sleep deprivation disrupts striatal anti-apoptotic responses in 6-hydroxy dopamine-lesioned parkinsonian rats

Objective(s): The present study was conducted to examine the effect of sleep deprivation (SD) on the anti-apoptotic pathways in Parkinsonian rats. Materials and Methods: Male Wistar rats (n = 40) were assigned to four groups (10 animals each): sham surgery (Sham), 6-hydroxydopamine (6-OHDA)-lesioned (OH), 6-OHDA-lesioned plus grid control (OH+GC), 6-OHDA-lesioned plus SD (OH+SD). Parkinson’s di...

Action of brain-derived neurotrophic factor on function and morphology of visual cortical neurons

Brain-derived neurotrophic factor (BDNF) is known to play a role in experience-dependent plasticity of the developing visual cortex. For example, BDNF acutely enhances long-term potentiation and blocks long-term depression in the visual cortex of young rats. Such acute actions of BDNF suggested to be mediated mainly through presynaptic mechanisms. A chronic application of BDNF to the visual cor...

BDNF–TrkB signaling in striatopallidal neurons controls inhibition of locomotor behavior

The physiology of brain-derived neurotrophic factor signaling in enkephalinergic striatopallidal neurons is poorly understood. Changes in cortical Bdnf expression levels, and/or impairment in brain-derived neurotrophic factor anterograde transport induced by mutant huntingtin (mHdh) are believed to cause striatopallidal neuron vulnerability in early-stage Huntington's disease. Although several ...

Effect of Endurance Training on Brain Derived Neurotrophic Factor (BDNF) and Tyrosine Kinase B (Trkb) Level in Hippocampus of Ischemic Induced Male Rats

Introduction: Brain derived neurotrophic factor (BDNF) have neuroprotective effect through binding with tyrosine kinase B (TrkB). Thus the Aim of the present study was to investigate the effects of eight weeks endurance training on BDNF and TrkB levels in the hippocampus of ischemic induced male rats. Methods: 40 Male wistar rats (12 weeks old and 228.19±21.18g) were divided into four groups, i...
